Mechanism of Action: Sildenafil acts by inhibiting phosphodiesterase type 5 (PDE5), an enzyme responsible for the degradation of cGMP in the corpus cavernosum. When PDE5 is inhibited, levels of cGMP increase, leading to smooth muscle relaxation and enhanced blood flow to the penis, thereby aiding in the establishment of an erection.
Applications:
- Erectile Dysfunction (ED): Primary use is in the treatment of ED.
- Pulmonary Arterial Hypertension (PAH): Used in lower doses to treat PAH.
Precautions: Can cause hypotension when taken with nitrates. Other side effects might include headache, dyspepsia, nasal congestion, impaired vision, photophobia, and blurred vision.
